The total protein band profile of the green leafhoppers (Nephotettix virescens) and of the rice (Oryza sativa) infected by tungro virus

Nusantara Bioscience 4: 32-35. Tungro virus is one of the important diseases of rice plants caused by double infection with RTBV and RTSV. One of the vectors is Nephotettix virescens Distant. The interaction between host and virus-vector still requires in-depth review. This study aimed: (i) to know the character of the total protein band profiles of rice plants infected with tungro virus and that of healthy rice plants, (ii) to know the character of the band profiles of total protein of N. virescens that consume the host rice plants infected by tungro virus and that of the healthy rice plants. Total protein band profiles of rice plants were identified using the electrophoresis method with SDS-PAGE. Sample extraction method employed buffer mercapto ethanol, while the sample extraction of green aphis employed buffer PBS IX. The painting of the protein bands employed the commasie blue ribbon. Data were analyzed descriptively based on the score of the migration of the band (Rf). The results showed that the protein content of 0.5 g of healthy leaves of rice plants was 0.567 g, whereas the protein content f 0.5 g of leaves of the rice plants infected by tungro virus was 1.011 g. The profile of protein bands on the tungro virus-infected rice plants was clearly thicker than that of the healthy rice plants. Protein bands with a higher quantity were expressed by the protein on the molecular weight of 108, and 117 kDa. Presumably these proteins are from the group of β-galactosidase, and bovine serum albumin. The function of such Proteins is still unknown, but presumably related to the plant’s responses to virus infection, because the protein did not appear in the healthy plants. The total protein content of green leafhoppers, which consumed the healthy leaves of rice plants was 0.1395 g, whereas the rice infected by tungro virus was 0.1546 g. Green leafhoppers, which consumed both healthy and tungro virus infected plants of rice showed no obvious differences on the protein expression, although it tended to be a bit thicker.

Abstrak. Sulistyarsi A, Suranto, Supriyadi. 2012. Profil pita protein total wereng hijau (Nephotettix virescens) dan inang tanaman padi (Oryza sativa) yang terinfeksi virus tungro. Nusantara Bioscience 4: 32-35. Virus tungro merupakan salah satu penyakit penting tanaman padi disebabkan oleh infeksi ganda RTBV dan RTSV dengan vektor antara lain, Nephotettix virescens Distant. Interaksi antara virus-inang dan vektor masih memerlukan kajian mendalam. Penelitian ini bertujuan: (i) mengetahui karakter profil pita protein total tanaman padi terinfeksi virus tungro dan tanaman padi sehat, (ii) mengetahui karakter profil pita protein total  N. virescens yang mengakuisisi inang tanaman padi terinfeksi virus tungro dan tanaman padi sehat. Profil pita protein total tanaman padi diidentifikasi menggunakan metode elektroforesis dengan SDS-PAGE. Metode ekstraksi sampel menggunakan buffer mercapto ethanol, sedangkan ekstraksi sampel wereng hijau menggunakan buffer PBS IX. Pengecatan pita protein menggunakan commasie blue. Data dianalisis secara deskriptif berdasarkan nilai migrasi pita (Rf). Hasil penelitian menunjukkan kadar protein 0,5 g daun tanaman padi sehat sebesar 0,567 μg, sedangkan kadar protein 0,5 g daun tanaman padi terinfeksi virus tungro sebesar 1,011 μg. Profil pita protein pada tanaman padi yang terinfeksi virus tungro jelas lebih tebal daripada pita protein di tanaman padi sehat. Pita protein dengan kuantitas lebih tinggi diekspresikan pada protein dengan berat molekul 108, dan 117 kDa. Diduga protein tersebut dari kelompok β-galaktosidase, dan bovine serum albumin. Fungsi protein tersebut belum diketahui, namun diduga ada kaitannya dengan respon tanaman terhadap infeksi virus, karena protein tersebut tidak muncul pada tanaman sehat. Kadar protein total wereng hijau yang mengkonsumsi daun tanaman padi sehat sebesar 0,1395 μg, sedangkan pada padi terinfeksi virus tungro sebesar 0,1546 μg. Wereng hijau, baik yang mengkonsumsi tanaman padi sehat maupun terinfeksi virus tungro, tidak menunjukkan perbedaan ekspresi protein jelas, meskipun cenderung lebih tebal.
